摘要
应用旋毛虫成虫期、新生幼虫期及肌幼虫期的排泄分泌物(ES)抗原作为包被抗原,ELISA方法检测感染旋毛虫试验小鼠2个月内不同天数血清中抗旋毛虫抗体Ig M和Ig G水平,并采用Excel软件绘制其抗体消长规律曲线并进行数据分析,了解旋毛虫感染宿主后不同发育时期刺激宿主机体产生抗旋毛虫抗体的变化规律,旨在为临床上能够合理利用旋毛虫不同发育时期的ES抗原诊断旋毛虫的感染提供理论依据。结果表明:可以利用旋毛虫3个发育时期ES抗原的混合物来检测旋毛虫45 d前的感染(即5~14 d的感染检测Ig M、15~45检测Ig G);感染45 d之后,可以利用单一肌幼虫时期ES抗原检测Ig G,最有意义的发现是利用旋毛虫成虫期和新生幼虫期的ES抗原可以有效检测14~19 d的感染。此研究成功获悉旋毛虫感染宿主后不同发育时期刺激机体产生抗旋毛虫抗体的消长规律,为进一步研究利用旋毛虫不同发育时期抗原诊断旋毛虫感染情况及解决"诊断盲区"(感染后14~19 d)问题提供了重要的科学依据。
Excretory-secretory antigen( ES) from adult stage( Ad3),newborn larvae( Ad6 +NBL) and muscle larvae( ML) of T. spiralis was used as coating antigen,tail vein blood of infected mice was collected on different days within two months,and serum was separated. On this basis,Ig M and Ig G anti-Trichinella was detected by ELISA method,and using Excel software,their antibody dynamic curves were drawn,followed by data analysis. The results indicated that the mixed ESantigen from the three developmental stages could be used to detect Trichinella infection before 45dpi( the serum anti-Trichinella Ig M was detected in the 5-14 dpi and anti-Trichinella Ig G was detected in the 15-45 dpi),the ML ES antigen could be used to detect Trichinella infection after 45 dpi,and the most significant finding was that AD3 and AD6 +NBL ES antigens could be used in 14-19 dpi. The study successfully obtained the dynamics of anti-Trichinella antibodies from the stimulated host by T. spiralis infection,which would provide important experimental foundation and theoretical reference for diagnosis on Trichinella spp.,infection and solution to "diagnostic gray zone "problem with different developmental stages' ES antigen from Trichinella spp.
