摘要
为有效提高猪血的利用开发率,采用液体发酵方法,以猪血水解度为测试指标,从屠宰场污水池中分离得到1株降解猪血蛋白能力较强的菌株X-17,并采用单因素试验和正交试验对X-17菌株培养基和发酵条件进行优化。获得其最佳培养基配方:玉米粉10 g/L,磷酸氢二钠1 g/L,硫酸镁0.5 g/L;最优发酵条件:接种量4%、培养温度37℃、培养基起始p H 7.0、发酵时间60 h。X-17通过最优条件发酵猪血后,水解度达18.52%,可溶性蛋白含量提高了181%,氨基态氮含量增加106%。
In order to effectively improve utilization of swine blood,by liquid fermentation method,using DH of fermented liquid as index,a strain X-17 with strong ability to degrade protein of swine blood was screened from the cesspool of a slaughterhouse. Culture medium and fermentation conditions of strain X-17 were optimized by single factor and orthogonal experiment. The optimal medium compositions obtained are: 10 g / L cornmeal,1 g / L Na2HPO4,0. 5 g /L Mg SO4,and the optimal fermentation conditionds are: amount of inoculation 4%,culture temperature 37 ℃,initial p H of culture medium 7. 0,fermentation time 60 h. Strain X-17 was fermented with the optimal conditions,degree of hydrolysis in fermentation liquid reached 18. 52%,and soluble protein increased by181% and amino nitrogen increased by 106%.
