载脂蛋白B基因rs2070665位点多态性与肃南裕固族、汉族人群血脂水平的相关性研究

Relationship between the polymorphism of ApoB gene rs2070665 loci and serum lipids in the Sunan Yugur and Han population

目的：探讨载脂蛋白B（ApoB）基因单核甘酸多态性rs2070665位点在甘肃裕固族、汉族人群中的分布及其与血脂水平的关系。方法：用高通量飞行质谱基因分型法（MALDI-TOF）分别对甘肃肃南裕固族227人和汉族306人进行ApoB基因rs2070665位点多态性检测和血脂水平检测,分析两者的相关性。结果：①裕固族血脂异常组总胆固醇（TC）和低密度脂蛋白胆固醇（LDL-C）水平明显高于汉族血脂异常组（P〈0.05）。②汉族TC和CC基因型分布频率明显高于裕固族（P〈0.01）;裕固族血脂异常组TC基因型LDL-C值高于汉族（P〈0.05）,裕固族血脂异常组CC基因型三酰甘油（TG）水平低于汉族（P〈0.05）,裕固族健康对照组TT基因型TG水平明显高于汉族（P〈0.01）;同民族内血脂异常组、健康对照组不同基因型血脂水平比较差异无统计学意义。③汉族T和C等位基因频率分布均高于裕固族（均P〈0.01）;同民族对比,裕固族ApoB基因rs2070665位点C等位基因携带者患血脂异常的风险高于T等位基因携带者（OR：1.778;95%CI：1.136-2.780）,汉族无差异。结论：甘肃肃南裕固族人群和汉族人群ApoB基因rs2070665位点基因多态性差异显著,裕固族人群ApoB基因rs2070665位点C等位基因携带者具有血脂异常遗传易感性。

Objective：To detect the association of the rs2070665 single nucleotide polymorphism（SNP）and serum lipids levels in the Sunan Yugur and Han population.Method：A total of 227 subjects of Yugur and 306 subjects of Han Chinese were included.DNA was extracted from venous blood with the method of phenol-chloroform.MALDI-TOF was used to analyze the apolipoprotein B gene polymorphism.The serum lipids were determined,and their relationships were analyzed.Result：①Total cholesterol（TC）and serum low-density lipoprotein cholesterol（LDL-C）were significantly higher in Yugur hyperlipidemia group than Han hyperlipidemia group.②The frequencies of TT,TC and CC genotypes had no difference in both ethnic hyperlipidemia groups,but the TC and CC genotype frequencies were significantly higher in Han than Yugur（P〈0.0125）;TC carriers in Yugur hyperlipidemia group showed higher serum LDL-C levels than that in Han hyperlipidemia group（P〈0.05）.CC carriers in Yugur hyperlipidemia group showed serum lower triglyceride（TG）levels than Han hyperlipidemia group（P〈0.05）.TT carriers in Yugur controls showed significantly higher serum TG levels than Han controls（P〈0.01）.Different genotypes of lipid levels in ethnic groups had no difference.③The allele frequencies of T and C had significant differnences in both ethnic groups（P〈0.01）;The risk of hyperlipidemia in C allele carriers in Yugur groups were higher than T allele carriers（OR：1.778;95%CI：1.136-2.780）.Conclusion：Yugur and Han people in Sunan area of Gansu province have significant differences in rs2070665 ApoB gene loci gene polymorphism,and C allele gene increased the risk of hyperlipemia in the Sunan Yugur.
