羟基红花黄色素通过RhoA/ROCK通路保护大鼠心肌再灌注损伤的机制研究 被引量:6

摘要 目的:研究羟基红花黄色素A对大鼠离体缺血心脏再灌注的保护作用,探讨其与Rho/ROCK信号通路的作用。方法:36只大鼠被随机分为7组,采用离体心脏灌流模型,给予缺血预处理及药物灌流,比较各组心率、冠状动脉流量,测定左心室内压,TTC心肌染色评估梗死范围,Western blot检测心肌组织内ROCK1、ROCK2活性以及Caspase-3的表达。结果:治疗组大鼠心脏缺血再灌注损伤减轻,心肌梗死面积减少,ROCK1、ROCK2活性以及Caspase-3的表达减低。结论:羟基红花黄色素A能够减轻大鼠心脏缺血再灌注损伤,同时减少心肌梗死面积,其作用机制可能与其抑制RhoA/ROCK信号通路,减少Caspase-3的表达有关。 Objective:To investigate the effects of hydroxysafflor yellow on rats' myocardial ischemia reperfusion and explore the relationship between hydroxysafflor yellow and the access of RhoA/ROCK.Method:Thirtysix rats were randomly divided into 7groups and made isolated heart perfusion model with the treatment of medical reperfusion.HR,CF,LV+dp/dtmax,LV-dp/dtmax,LVDP were detected.The myocardial infarct size was detected by TTC,and the density of ROCK1,ROCK2 and Caspase-3were detected by Western blot.Result:The damage of myocardial ischemia reperfusion injury and the expression of ROCK1,ROCK2 and Caspase-3were reduced in treatment group.Conclusion:Hydroxysafflor yellow can protect the myocardial ischemia reperfusion injury in rats,and also reduce the infarct size.The mechanism may be related to inhibition of the signalling pathway of RhoA/ROCK,and the reduction of Caspase-3expression.
