氧化还原内稳态关键因子Nrf2对新陈代谢的调节作用

Regulation of Metabolism by Transcription Factor Nrf2 via Redox Homeostasis

摘要 核转录因子红细胞系2-p45相关因子2(nuclear factor-erythroid 2-p45 related factor 2,Nrf2)是调控细胞氧化还原稳态的重要转录因子。除了调节氧化应激反应外,近年来,Nrf2在代谢调节中的重要作用也备受关注。文章重点综述了Nrf2调控细胞的糖代谢、脂代谢、核酸代谢和氨基酸代谢的方式,并论述了Nrf2失调导致的多种代谢相关疾病,包括糖尿病、脂肪肝、帕金森氏症和肿瘤。最后,讨论了代谢压力介导的Nrf2活性改变与长寿、肿瘤防治及压力应激的关系。 Nuclear factor-erythroid 2 p45-related factor 2 (Nrf2) is an important transcription factor regulating the homeostasis of cellular redox, in addition to its role in the response to oxidative stress, Nrf2 attracts much attention as the key factor in the regulation of metabolism. This review mainly summarizes the mechanism by which Nrf2 controls the metabolism of sugar, lipid, nuclear acid and amino acid, and discusses the metabolism-related diseases caused by Nrf2 deregulation, including diabetes, fatty liver, Parkinson's disease and cancers. At last, the associations of the alteration in Nrf2 activity mediated by metabolic stress with longevity, cancer prevention and stress response are also discussed.
