摘要
选取‘红城7号’甜瓜的子叶为外植体,接种于含有不同激素配比的MS培养基中,进行了组织培养研究。结果表明:‘红城7号’甜瓜叶片离体培养的最佳愈伤组织诱导培养基为MS+6-BA 2.00 mg·L-1+2,4-D 0.10 mg·L-1,诱导率达90.0%,在MS+6-BA 2.00 mg·L-1+2,4-D 0.01 mg·L-1培养基中的出芽率最好,分化率为86.7%。在MS+IBA 0.50 mg·L-1生根培养基上诱导不定芽生根的效果最好,生根率达83.3%。试验为‘红城7号’甜瓜再生植株的进一步研究提供了理论依据。
The cotyledons of‘Hongcheng NO.7'melon were selected as explants and planted into the MS medium with different concentration, its tissue culture were studied. The research results showed that the most ideal medium for callus inducing was MS+6-BA 2.00 mg· L-1 +2,4-D 0.10 mg-L-lwith the best induction rate 90.0%;The most ideal medium for adventitious buds inducing was MS + 6-BA 2.00 mg.L-1 + 2,4-D 0.01 mg·L-1 with the induction rate 86.7%; The best rooting medium was MS+ IBA 0.50 mg·L-1 with the best rooting rate 83.3%. The study could provide a experimental basis for further study on the plant regeneration in the ‘ Hongcheng NO.7' melon.
