棒状简枝霉YX016产蒽醌发酵条件的优化研究

Optimization on fermentation conditions of Simplicillium obclavatum YX016 for the production of anthraquinones

摘要 目的:从竹黄中分离筛选到可以发酵产生蒽醌的一株棒状简枝霉YX016,为提高蒽醌产量,对液体发酵条件进行优化。方法:通过单因素实验选出合适的碳源、氮源、微量矿质元素和生长因子等,再以葡萄糖、尿素、KH2PO4、Mg SO4、H3BO3、生长因子VB1和初始p H进行7因素4水平的正交优化实验。结果:确定的最佳培养基配方为葡萄糖30 g/L、尿素0.5 g/L、KH2PO4 0.2 g/L、Mg SO4 0.05 g/L、H3BO3 0.08 g/L、VB1 21μg/100 mL,初始p H6.5,装液量为100 mL/250 mL,接种菌龄为5 d,发酵温度为27℃,摇床转速为150 r/min。在优化的发酵条件下,蒽醌的产量达到69.05μg/m L,比优化前提高了1.95倍。 Objective:A strain of producing anthraquinones,Simplicillium obclavatum YX016,was isolated from the fruiting body of Shiraia bambusicola.The liquid culture conditions for anthraquinones were optimized in order to enhance the production of anthraquinones.Methods:The effects of carbon source,nitrogen source,trace mineral elements,growth factors and rotatory speed on the production of anthraquinones was researched by means of single-factor test.Furthermore,the orthogonal test which contained seven factors and four levels was employed to evaluate the effects of glucose,urea,KH_2HPO_4,MgSO_4,H_3BO_3,V_(B1) and initial pH on the production of anthraquinones.Results:The optimal medium formulation was determined as follows:glucose 30 g/L,urea 0.5 g/L,K_2HPO_4 0.2 g/L,MgSO_4 0.05 g/L,H_3BO_3 0.08 g/L,V_(B1) 21 μg/100 mL,initial pH 6.5,medium volume 100 mL was in 250 mL flask,inoculant age of 5 d,incubation temperature 27 ℃,rotatory speed 150 r/min.Under the optimal conditions,the yield of anthraquinones could reach 69.05 μg/mL,which was 1.95 folds compared to that in the basic culture medium.
