摘要
巨大口蘑是一种珍稀食用菌,具有抗褐变特性,了解其褐变相关酶的特性,对探明其抗褐变机理具有重要意义。采用不同金属离子(Zn^2+、Mg^2+、Cd^2+、Cu^2+、Fe^2+、Fe^3+、Mn^2+)的梯度溶液浸泡法处理蘑菇切片,并测定了不同金属离子对其褐变相关酶活力影响。结果表明:Cu^2+、Zn^2+、Mg^2+对其多酚氧化酶(PPO)具有显著低促高抑作用,Fe^3+为激活作用,Fe^2+和Cd^2+起抑制效果。0.4-2 mmol/L的Zn^2+、Mg^2+和Cd^2+对其酪氨酸酶(TYE)有显著激活作用,Cu^2+具有低促高抑作用;Fe^3+、Fe^2+和Mn^2+对TYE活力影响不显著。Cd^2+、Cu^2+、Fe^3+、Mn^2+和Zn^2+对其苯丙氨酸解氨酶(PAL)有显著激活效果,Fe^2+对PAL有抑制作用;Mg^2+对PAL无显著影响。
In order to explore the mechanism of Trichoioma giganteum anti-browning,the gradient solution immersion method with different metal ions(Zn^2+,Mg^2+,Cd^2+,Cu^2+,Fe^2+,Fe^3+,Mn^2+) was adopted to process mushroom slices,and the influence of different metal ions on the browning related enzyme activity was determinated in this paper.The results showed that Cu^2+,Zn^2+,Mg^2+ had significant stimulation on the poiyphenoi oxidase(PPO) activity at low concentration and had inhibition at high concentration.Fe^3+ could active PPO activity.Fe^2+ and Cd^2+ had inhibitory effect.0.4-2 mmol/L Zn^2+,Mg^2+ and Cd^2+ had significant activation on the tyrosinase(TYE) activity.Cu^2+ had stimulation at low concentration and had inhibition at high concentration.The influence of Fe^3+,Fe^2+ and Mn^2+ on the TYE activity was not significant.Cd^2+,Cu^2+,Fe^3+,Mn^2+ and Zn^2+ had significant activation on the phenylalanine ammonialyase(PAL) activity.Fe^2+ had inhibition on PAL;Mg^2+ had no significant influence on PAL activity.
