摘要
目的:应用基因芯片技术研究异紫堇碱处理后的Si Ha细胞差异基因表达谱改变,以寻找异紫堇碱抑制宫颈癌Si Ha细胞HPV基因表达的分子机理。方法:抽提经异紫堇碱处理后不同时段的Si Ha细胞和未经异紫堇碱处理的Si Ha细胞中的mRNA来制备探针,经杂交、洗涤后,通过计算机扫描分析未经异紫堇碱处理的Si Ha细胞和处理后的Si Ha细胞基因表达谱的差异情况。结果:在每组60000多个候选基因中,各筛选出200多条差异表达倍数在2倍以上的基因,在异紫堇碱处理后12小时的样本中下调基因206条,上调基因70条,未知基因217条;24小时后下调基因95条,上调基因137条,未知基因166条;48小时后下调基因150条,上调基因101条,未知基因159条。结论:基于基因芯片技术的基因表达谱分析能够高通量筛选与异紫堇碱抑制宫颈癌Si Ha细胞HPV基因表达的分子机理相关的基因。
Objective: Make an analysis of diferential gene expression profiles in Isoeorydine-treated SiHa cell with eDNA microarray to find out the molecular mechanism that the Isoeorydine inhibit the expression HPV in SiHa cell base on. Methods: Total RNA was extracted from the Iso- corydine-treated SiHa cell at different time and normal SiHa cell. The mRNA was used to prepare probes. The mixed probes were hybridized to the eDNA mieroarray. After washing, the DNA chips were scanned by laser scanner, The acquired was analyzed. Results: Among the more than 60000 target genes of the 3 groups,more than 200 genes were identified in Isoeorydine-treated SiHa cells that were either over or under- expressed as compared to controls in each group. And about two-thirds were found in this study. Conclusion:The eDNA mieroarray for analy- sis of gene expression profile is a powerful method for finding out the meeular meehanism that the lsoeorydine inhibit the expression HPV in SiHa cell base on.
