摘要
目的:探讨毛蕊花糖苷对内毒素(lipopolysaccharide,LPS)诱导小鼠的急性肺损伤(acute lung injury,ALI)保护作用。方法:于小鼠腹腔注射LPS(8 mg/kg)复制ALI动物模型。将小鼠随机分为空白组对照组、LPS组、地塞米松组(2 mg/kg)、毛蕊花糖苷组(20 mg/kg、40 mg/kg)。观察各组肺组织病理学改变,测量肺湿/干重比,支气管肺泡灌洗液中性粒细胞百分比、超氧化歧化酶(SOD)、丙二醛(MDA)、炎症因子白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、前列素E2(PGE2)含量及肺组织中环氧酶-2(COX-2)5-脂氧合酶(5-LOX)蛋白表达。结果:毛蕊花糖苷组(20 mg/kg、40 mg/kg)可有效减轻LPS所致肺组织病理学变化,能降低肺泡灌洗液中炎症细胞的数目、MDA、IL-6、TNF-α、PGE2,增加肺泡灌洗液中SOD;著降低肺湿/干重比重,减低COX-2与5-LOX蛋白水平。结论:毛蕊花糖苷20 mg/kg、40 mg/kg可减轻LPS所致急性肺组织损伤,对LPS诱导的急性肺损伤有保护作用。
Objective: To determine the protective effect of acteoside on acute lung injury induced by lipopolysaccharlde (LPS). Methods: The BABL/C mice were randomly allocated into control, LPS, dexamethasone (2mg/kg), acteoside (20 mg/kg), and acteoside (40 mg/kg). The bronchoalveolar lavage fluid (BALF) samples were analyzed for the concentration of numbers of neutrophilic, SOD, MDA, prostaglandin E2 (PGE2), interleukin-6 (IL-6) and tumor necrosis factor alpha (TNF-ct), and The lung samples were taken for histologlc evaluation and for determination of wet-to-dry(W/D) lung weight, The protein levels of cyclooxygenase-2 (COX-2) and 5-1ipoxygenase were detected by Western blot analysis. Results: Pretreatment with acteoside prior to the administration of intratracheal LPS significantly induced a decrease in lung wet weight/dry weight ratio, in total leukocyte number and neutrophil percent in the BALF, and in PGE2 of lung in dose-dependent man- ners, and in IL-6, the tumor neurosis factor-or (TNF-ct) in the BALF and COX-2 and 5-LOX proteins in lung. These results showed that an- ti-inflammatory effects of acteoside against the LPS-induced ALI.
