摘要
目的:通过建立大鼠血管性痴呆模型,研究山茱萸多糖对血管性痴呆大鼠学习记忆、脑组织氧化应激反应的影响。方法:将雄性SD大鼠60只,随机分为6组,包括假手术组、模型组、山茱萸多糖0.14g/kg、0.28g/kg、0.56g/kg组和盐酸多奈哌齐0.52mg/kg组。除假手术组外,其余各组均制备血管性痴呆模型,其中假手术组与模型组每天灌胃生理盐水,其余各组分别连续28天灌胃不同剂量山茱萸多糖和盐酸多奈哌齐。28天后,分别对6组大鼠行morris水迷宫测试,测试完成后,断头取脑,测定脑组织中单胺氧化酶(MAO)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)含量。结果:morris水迷宫测试结果显示,随着训练时间延长各组大鼠逃避潜伏期时间逐渐变短,0.28g/kg、0.56g/kg山茱萸多糖组大鼠平均逃避潜伏期明显缩短;空间探索试验中,与模型组比较,0.28g/kg、0.56g/kg山茱萸多糖组大鼠在平台象限路径百分比与平台象限停留时间明显长于模型组。与假手术组相比,模型组脑组织GSH-Px、CAT活性明显下降,MAO活性明显升高;与模型组相比,0.28g/kg、0.56g/kg山茱萸多糖组大鼠脑组织GSH-Px、CAT活性明显升高,MAO活性明显下降。结论:山茱萸多糖能够明显提高血管性痴呆大鼠学习记忆能力,其作用机制可能通过对抗机体氧化应激。
Objective: A rat model of vascular dementia was established in order to investigate the functional mechanism and the effect of the learn- ing and memory and oxidative stress of the brain on the Comus officinalis polysaccharide. Methods: Sixty male rats were randomly divided into six groups : sham operated group, model group, dopenezil,0.14g/kg,0.28 g/kg,0.56 g/kg Comus oitlcinalis polysaccharide group. The other groups were all established VD model except for the sham operated group. The sham operated group and the model group were lavaged with saline for 28 days and the other groups were lavaged with dopenezil and different doses of Comus officinalis polysaccharide for 28 days. After 28 days all the rats were tested by morris water maze. And then,the activity of MAO,GSH-Px,CAT were measured in the brain. Results:The morris water maze test showed that the latency of escape among groups were shorter and shorter. The 0.28g/kg,0.56g/kg Comus officinalis polysaccharide could decrease the latency of escape; In spatial probe test the time spent in target quadrant and the percentage of the distance of the model group were obviously increased as compared with the 0.28 g/kg,0.56g/kg Comus officinalis polysaccharide group ( P 〈0.05 or P 〈 0.01 ). The activity of GSH-Px,CAT of the model group was much lower than the sham operated group ( P 〈 0.01 ), but the activity of MAO was opposite (P 〈 0.01 ) ;The activity of GSH-Px, CAT of the middle, high dose of Comus otlicinalis polysaccharide group was higher than the model group ( P 〈 0.05 or P 〈 0.01 ), but the activity of MAO was opposite ( P 〈 0.05 or P 〈 0.01 ). Conclusion: The Comus offici- nalis polysaccharide could obviously improve the ability of the learning and memory. And the mechanism may be that it prevented from the oxi- dative stress of the brain.
