补骨脂素对HepG2细胞BSEP、NTCP的影响 被引量:12

Study on the influence of Psoralen on HepG2 cell proliferation in vitro and the expression of BSEP,NTCP Proteins

摘要 目的观察补骨脂素对人肝癌细胞HepG2的体外细胞毒性,并研究其可能的机制。方法采用MTT法观察补骨脂素对HepG2细胞增殖的影响,考察其体外毒性。运用Western印迹法、ELISA试剂盒检测补骨脂素作用HepG2细胞24h后胆盐输出泵(BSEP)、Na+胆盐共转运体(NTCP)蛋白的表达。结果补骨脂素对HepG2细胞有体外细胞毒性。药物作用24h补骨脂素的IC50为176.5813μmol/L,补骨脂素能够抑制BSEP蛋白的表达,促进NTCP蛋白的表达。结论补骨脂素对HepG2有体外细胞毒性,且这种毒性可能与其影响了胆汁酸转运体有关。 Objective Study the cytotoxicity of Psoralen on human hepatocellular carcinoma HepG2 cell in vitro,to explore its possible mechanism.Methods Observe the effect of Psoralen on HepG2 cell proliferation through MTT to make clear of toxicity in vitro.Detect the expression of bile salt export pump( BSEP),Na+- taurocholate cotransporting polypeptide( NTCP) protein after Psoralen working HepG2 cells for 24 hours by using Western Blot and ELISA kit.Results Psoralen has cytotoxicity on HepG2 cell in vitro.The IC50 of Psoralen were 176.58 μmol / L.Psoralen inhibited BSEP protein and enhanced the expression of NTCP protein.Conclusion The toxicity of Psoralen on HepG2 cell in vitro may be related to its influence on bile acid transporter.
