大麻素Ⅱ型受体激动剂对A375细胞增殖的影响 被引量:3

EFFECT OF CANNABINOID RECEPTOR Ⅱ AGONIST ON THE PROLIFERATION IN A375 CELL LINE

摘要 目的观察大麻素Ⅱ型受体激动剂JWH-133和WIN55,212-2对A375细胞增殖的抑制作用。方法以人恶性黑素瘤A375细胞和人永生化角质形成细胞株HaCat为体外实验模型,RT-PCR检测CB-R2在A375细胞中mRNA水平的表达;不同剂量大麻素Ⅱ型受体激动剂JWH-133和WIN55,212-2处理组。MTT法测定大麻素Ⅱ型受体激动剂JWH-133和WIN55,212-2对A375细胞增殖的影响。结果 1人恶性黑素瘤A375细胞和人永生化角质形成细胞株HaCat在mRNA水平均表达CB-R2;2不同浓度(0.01,0.1,1,10,100μmol/L)大麻素Ⅱ型受体激动剂JWH-133和WIN55,212-2抑制A375细胞增殖,抑制作用48h达高峰;3比较大麻素Ⅱ型受体激动剂JWH-133和WIN55,212-2两种药物对A375细胞增殖的抑制作用差异无统计学意义(P>0.05)。结论 1A375细胞在mRNA水平表达CB-R2;2JWH-133和WIN55,212-2抑制A375细胞增殖。 Objective To investigate the effect of Cannabinoid receptor JWH-133 and WIN55,212-2 on the proliferation in A375 cells in vitro.Methods The expression of CB-R2 in A375 cells and HaCat cells was detected by RT-PCR for exploring the inhibitory effect of CB-R2 agonist in antiproliferation on A375 cells.Malignant melanoma A375 cells and HaCat cells were cultured under different doses of JWH-133 and WIN55,212-2 agonist and solvent controls.The effect of JWH-133 and WIN55,212-2 on cell proliferation was observed.The growth rate of the cells was detected by MTT assay.Results A375 cells and HaCat cells had expressed the CB-R2 in mRNA levels.Different concentrations(0.01/0.1/1/10/100umol/L)of JWH-133 and WIN55,212-2 played an effect(P〈0.05)on the inhibition of A375 cells,the peak time was at 48h(P〈0.05).No significantly difference between JWH-133 and WIN55,212-2on inhibition of proliferation of A375 cells(P〉0.05).Conclusions 1A375 cells has expressed the CB-R2 in mRNA Levels;2JWH-133 and WIN55,212-2 siginfficantly inhibit A375 cells proliferation.
