摘要
为了有效保护和利用野生华北蓝盆花(Scabiosa tschiliensis Grunning)资源,通过离体培养技术建立了华北蓝盆花再生体系.研究结果表明:以子叶为外植体,诱导愈伤组织较适宜的培养基为MS+TDZ1.0mg/L,诱导率达82.74%;诱导不定芽的培养基为MS+6-BA 4.0mg/L,诱导率为86.30%;生根培养基为MS+IBA 2.0mg/L,生根率为73.85%.愈伤组织、野生植株总黄酮含量较高,其醇提物具有较强的抗氧化活性,对DPPH自由基具有很高的清除率.
For protection and utilization of the wild Scabiosa tschiliensis Grunning,apropagation system in vitro was established by using leaf explants from aseptic seedlings of S.tschiliensis.The results showed that the optimal medium for callus induction was MS+TDZ 1.0mg/L,and induction rate was about 82.74%.Adventitious shoots developed when explants were cultured on MS+6-BA 4.0 mg/L and induction frequency was as high as 86.30%.MS+IBA 2.0 mg/L was the best medium for rooting and73.85% of the regenerated shoots formed complete plantlets.Quantitative analysis of flavonoids showed that the phytochemical profiles of calluses were similar to that of wild-type plants.The 2,2-diphenyl-1-picrylhydrazyl(DPPH)assay revealed that the flavonoid extracts of calluses and wild-type plants had strong antioxidant activities.
