摘要
目的研究碱基切除修复酶OGG1对PM2.5造成肺上皮细胞线粒体损伤的保护作用。方法采集湛江市附属医院附近的PM2.5颗粒物,培养肺上皮细胞beas-2b,并转染OGG1过表达载体,以PM2.5 100μg/ml 24 h分别染毒正常和OGG1过表达的beas-2b细胞,DCFH染色后流式测定细胞内活性氧水平,单细胞凝胶电泳检测DNA损伤水平。结果OGG1过表达后能显著抑制PM2.5造成的细胞内活性氧水平的升高,并能显著抑制PM2.5造成的beas-2b细胞DNA损伤。结论 OGG1能修复PM2.5造成的DNA损伤,该作用可能与其抑制ROS作用相关。
Objective To explore the repair mechanism of OGG1 on mitochondrial damage caused by PM2.5.Methods PM2.5 near the Affiliated Hospital of Guangdong Medical College was collected.Human bronchial epithelial cell line (BEAS-2B) transfected with OGG1 overexpression vector were exposed to the PM2.5(100 μg/ml) for 24h.The level of intracellular reactive oxy-gen species was assessed by DCFH-DA staining.The DNA strand breaks were assessed by comet assay.Results OGG1 could in-hibit the generation of reactive oxygen species induced by PM2.5.The OGG1 could alleviate DNA damage caused by PM2.5.Con-clusion The base excision repair enzymes OGG1 alleviate DNA damage and this effect may be related to the role of ROS inhibition .
