摘要
目的:分析培养基、外源激素和真菌诱导子对北柴胡毛状根生长和柴胡皂苷含量的影响,筛选出柴胡皂苷产量高的培养条件。方法:设置3种培养基(B5、改良White和WPM)、3种外源激素(IBA、IAA和NAA)、真菌诱导子为黑曲霉诱导子,每组处理设计2~4个浓度梯度,25℃、110 r/min暗培养,3个月后收获称重,测定根产量,HPLC方法检测毛状根柴胡皂苷含量。结果:在添加0.5 mg/L IBA的B5培养基中,毛状根和柴胡皂苷产量最高,分别为2.4 g和9.05mg。结论:筛选出北柴胡毛状根最适培养条件,为北柴胡毛状根大规模培养利用提供了参考依据。
Objective:To study the effects of different media,phytohormones and fungus elicitors on the growthand saikosaponin production of Bupleurum chinense DC.hairy roots.Methods:Conditions were screened includingthree different culture media(B5,modified-White and WPM),three types of exogenous hormones(IBA,IAA andNAA) and fugus elicitor from Aspergillus niger.Two to four concentration gradients were set for each group.Theculture condition was at 25℃,110 r/min.After three months,the weights of roots were measured and the saikosa-ponin content were determined by HPLC method.Results:The production of roots and saikosaponin were highestin the B5 culture medium added with 0.5 mg/L IBA,those were 2.4 g and 9.05 mg,respectively.Conclusion:Theoptimal condition for saikosaponin production from hair roots which we screened out will provide a basis for fur-ther cultivation in large scales.
