地黄梓醇对原代培养SD乳大鼠成骨细胞成骨活性的影响研究 被引量:15

摘要 目的观察不同浓度地黄梓醇对体外培养新生SD大鼠成骨细胞增殖及成骨活性的影响,为地黄防治骨质疏松症提供实验依据。方法体外培养原代新生24 h SD大鼠成骨细胞,细胞传至第5代,分别加入1×10-3,1×10-5,1×10-7,1×10-9mol/L梓醇溶液及生理盐水10μL/孔,继续培养24,48,72 h后,分别检测成骨细胞增殖情况、骨钙素(BGP)及碱性磷酸酶(ALP)活性。结果培养24 h时,1×10-3mol/L梓醇溶液能显著促进成骨细胞增殖(P<0.05);培养48 h时,1×10-3mol/L梓醇溶液促增殖作用最强(P<0.05)。培养48h时,1×10-7mol/L组和1×10-3mol/L组ALP活性显著高于对照组(P均<0.05);培养72 h时,1×10-3mol/L组ALP活性显著增加(P<0.05)。培养48,72 h后,各浓度梓醇溶液组BGP活性均高于对照组(P均<0.05),且1×10-3mol/L组明显高于其他浓度组(P均<0.05)。结论梓醇在1×10-5mol/L和1×10-3mol/L范围对体外培养SD大鼠成骨细胞的增殖、分化及成骨作用相对显著,尤其在1×10-3mol/L浓度下作用最显著,且具有时效及量效关系。 Objective It is to observe the effect of Catalpol at different concentrations on proliferation and osteoblast activity of primary cultured osteoblast in neonatal SD rats,thus to provide experimental evidence for the prevention and treatment of osteoporosis. Methods Primary rat neonatal 24 h SD osteoblasts were cultured in In vitro,when the cells spread to the 5th generation,they were added with different concentrations of 1 × 10-3,1 × 10-5,1 × 10-7,1 × 10-9mol / L catalpol saline solution 10 μL / hole,then the culture continued,in 24 h,48 h,72 h after culture the proliferation,ALP activity and activity levels of BGP of osteoblasts were detected. Results When cultured for 24 h time,1 × 10-3mol / L Catalpol could promote the proliferation of osteoblasts significantly( P〈0. 05); For 48 h,the proliferation effect of 1 × 10-3mol / L Catalpol was the strongest( P〈0. 05). Compared with the control group when cultured for 48 h,the activity of ALP in 1 × 10-7mol / L and 1 ×10-3mol / L was significantly higher( P〈0. 05),for 72 h in 1 × 10-3mol / L group,the ALP activity was increased significantly( P〈0. 05). When cultured for 48 h and 72 h,compared with the control group,the activity of BGP in every Catalpol group was all higher( all P〈0. 05),especially in 1 × 10-3mol / L group,the activity was the highest than the other groups( P〈0. 05). Conclusion Catalpol at the concentration range from 1 × 10-5mol / L to 1 × 10-3mol / L had significant effect on the proliferation,differentiation and osteoblast activity of primary cultured osteoblast in vitro in neonatal SD rats,especially the effect at concentration of 1 × 10-3mol / L was most significant,and there was relationship of time-effect and dose-effect.
