摘要
目的探讨分析多西他赛化疗前后白细胞介素-8(interleukin-8,IL-8)在乳腺癌间质成纤维细胞中的表达变化。方法采集青岛市立医院乳腺外科2014-03-01-05-31未经过新辅助化疗的乳腺癌患者手术切除标本10例,通过原代培养建立稳定的乳腺癌间质成纤维细胞株。每一株原代细胞传代后分为两组。一组不做特殊处理,标记为对照组;另外一组经20ng/mL多西他赛(泰索帝)处理24h,标记为实验组。分别提取两组细胞的mRNA,应用Illumina光纤微珠芯片平台进行基因表达谱分析并筛选差异基因。继而通过Real-time PCR及蛋白质印迹法检测IL-8在两组乳腺癌间质成纤维细胞中的表达变化。结果多西他赛化疗后乳腺癌间质成纤维细胞的基因表达谱发生变化,包括17例上调表达基因及18例下调表达基因(含IL-8)。通过实时定量PCR及蛋白质印迹法检测发现,多西他赛化疗后乳腺癌间质成纤维细胞中的IL-8基因及蛋白表达量高于化疗前,差异有统计学意义,t=-4.274,P=0.013。结论多西他赛化疗后乳腺癌间质成纤维细胞中IL-8表达上调,提示其可能与化疗疗效及耐药相关。
OBJECTIVE To explore the expression of interleukin-8 (IL-8) in the carcinoma-associated fibroblasts (CAFs) of breast cancer before and after chemotherapy. METHODS The tissue specimens from breast cancer patients who underwent complete surgical resection were collected for primary cultures of carcinoma-associated fibroblasts. Each pair of CAFs was obtained from the same patient and classified into two groups. One group was treated with 20 ng/mL Taxotere for 24 h (regarded as after chemotherapy) while the other group was not processed with Taxotere (regarded as before chemotherapy). Microarray studies were performed using illumina Beadarray, to analyze the gene expression pro- file of CAFs before and after chemotherapy and to select candidate genes. Real-time PCR and Western-blot assays were performed to detect the expression of IL-8 in CAFs of breast cancer before and after chemotherapy. RESULTS 17 up- regulated genes and 18 down-regulated genes were identified through the gene expression profiling of CAFs in breast canc- er before and after chemotherapy,including IL-8. The CAFs after chemotherapy showed higher gene and protein expres- sion of IL-8, compared to the group before treatment (For Real-time PCR,t= -4. 274,P=0. 013). CONCLUSION The high expression of IL-8 may associate with chemotherapy efficiency and resistance.
