多烯磷脂酰胆碱对放射性肺损伤大鼠MCP-1和TGF-β1表达的影响

摘要 目的:探讨多烯磷脂酰胆碱(PPC)对放射性肺损伤大鼠TNF-α和转化生长因子-β1(TGF-β1)表达水平的影响。方法:单次剂量30Gy照射大鼠右肺建立放射性肺损伤模型。96只大鼠随机分为4组:空白对照组(A组)、PPC照射组(B组)、PPC佯装组(C组)及生理盐水(NS)佯装组(D组),每组24只。A,B组于照射后分别予NS和PPC腹腔注射1周;C、D组于佯装照射后分别予PPC和NS腹腔注射1周。照射后第7,30,90天后采集血标本并离心取血清,采用酶联免疫吸附测定法(ELISA)检测血清中TGF-β1和单核细胞趋化蛋白-1(MCP-1)的含量。结果:A组在放疗后3个时间点的血清TGF-β1和MCP-1水平均明显高于D组(P<0.05),B组在放疗后3个时间点的血清TGF-β1和MCP-1水平均明显高于C组(P<0.05),并且B组大鼠血清TGF-β1水平在放疗后30天明显低于A组(P<0.05),而血清MCP-1水平则在放疗后第7天明显低于A组(P<0.05)。结论:PPC可降低放射性肺损伤大鼠血清TGF-β1和MCP-1的表达水平。 Objective: Toinvestigate the effects of polyene phosphatidylcholine(PPC) on the expression of transforming growth factor-β1 (TGF-β1)and monocyte ehemotactic protein-1 (MCP-1) in rats with radiation-induced pulmonary injury. Methods: The radiation-induced right lung injury rat model were established by a single dose of 30 Gy. 96 SPF male SD rats(6-8 weeks old,220-250 g) were randomly and equally divided into 4 groups: normal saline group(A group),PPC irradiated group (B group),PPC sham group (C group),NS sham irradiated group (D group),every group had 24 rats. Group A and B were respectively received intraperitoneal injection with normal saline(NS) and PPC for a week. Group C and D respectively were administered intraperitioneally with PPC and NS for 1 week after sham irradiation, too. Then, the blood samples from these rats were collected at pointed day,namely irradiation after the 7th day, the 30th day and the 90th day so that centrifuged to obtain serum. Last, ELISA method was used to detect the expression of TGF-β1 and MCP-1 in serum. Results: The serumal level of TGF-β1 and MCP-1 of group A at the 3 time points were both significantly higher than that in group D ( P 〈0.05). Also, the level of TGF-β1 and MCP-1 in group B at the 3 time points were significantly higher than that in group C ( P 〈0.05). Radiotherapy after 30 days, the serum level of TGF-β1 of group B was significantly lower than that of group A ( P 〈0.05) ,while the levels of MCP-1 in group B were significantly lower than that of group A when radiotherapy after 7 days, ( P 〈0.05). Conclusion: PPC can decrease the serumal expression of TGF- β1 and MCP-1 in rats with radioactive lung injury.
