Expression and significance of C-C chemokine receptor 2 in nonalcoholic steatohepatitis

目的初步探讨CCR2在NASH患者中的表达及意义。方法收集NASH患者肝穿标本15例,健康对照肝组织标本5例,进行HE染色及NAS评分、免疫组化染色、激光共聚焦染色等,检测巨噬细胞、CCR2的表达情况并分析与疾病严重程度的相关性。结果 NASH患者肝组织中巨噬细胞表达较健康对照组明显增加,并与NAS评分呈正相关。NASH患者肝组织中CCR2表达较健康对照组明显增加,并且主要表达在肝内巨噬细胞表面。结论 CCR2可能通过肝内巨噬细胞参与NASH的发展。

Objective To investigate the expression and significance of CCR2 in patients with NASH preliminarily. Methods Fifty specimens of liver tissue from patients with NASH and 5 from healthy controls were collected.The tissues were stained with H＆amp;E to estimate the NAFLD activity score （NAS）.Detection of macrophages and expression of CCR2 were assessed by immunohistochemistry to analyze the correlation with the degrees of NASH.Moreover,the distribution of macrophage and CCR2 were observed by confocal laser scanning.Results The expression of macrophages was significantly increased in NASH patients than that in healthy controls,which positively correlated with NAFLD activity score.NASH patients also had significantly higher CCR2 levels than the controls,which mainly expressed on the surface of hepatic macrophages.Conclusion CCR2 probably promotes NASH progression through macrophages.
