二甲双胍对糖尿病小鼠血管内皮细胞氧化应激的影响 被引量:1

摘要 目的:探讨二甲双胍对糖尿病模型小鼠血管内皮细胞中氧化应激的影响。方法:雄性小鼠(30只)随机分为正常对照组(枸椽酸钠缓冲液注射)、糖尿病模型组(链脲菌素注射)和二甲双胍模型组(链脲菌素注射+二甲双胍口服),每组10只。链脲菌素(80 mg·kg-1·d-1)诱导造模成功后二甲双胍模型组予二甲双胍(50 mg·kg-1·d-1),正常对照和糖尿病模型组正常饮水,定期称重,并检测各阶段血糖。1周后处死小鼠,取股直肌免疫组化染色,观察毛细血管内皮细胞氧化应激状态。结果:与糖尿病模型组比,二甲双胍模型组血糖降低,体质量增加(P<0.05)。免疫组化染色示小鼠肌间隙毛细血管内皮细胞氧化应激标志物水平均明显降低。结论:二甲双胍可缓解血管内皮细胞氧化应激,对糖尿病血管并发症的发生具有预防作用。 Objective:To evaluate the effect of metformin on oxidative stress in vascular endothelial cells from diabetic model mice.Methods:Thirty male mice were randomly divided into normal control(treated with citron acid sodium buffer),diabetic model(treated with streptozotocin)and metformin model(treated with streptozotocin and metformin)groups,10 in each group. The mouse model was established by intraperitoneal injection of streptozotocin(80 mg·kg-1·d-1). The metformin group were given 50 mg·kg-1·d-1,and other two groups were given normal drinking water.The body mass was determined before and after treatment.The immunohistochemistry analysis was performed to observe the level of oxidative stress markers in vascular endothelial cells in rectus femoris. Results:Compared with the diabetic model group,the metformin model group had significantly lower level of blood glucose,higher body weight(P0.05). Conclusion:Metformin can relieve oxidative stress in vascular endothelial cells and may prevent vascular complication of diabetes.
