稳定表达重组人CD317细胞系的构建及其对流感病毒的抗性研究

摘要 目的:研究表达重组人CD317细胞系对流感病毒的抵抗作用。方法:将CD317基因真核表达质粒转染MDCK细胞,用含有G418培养基进行筛选,挑取抗性细胞克隆扩大培养,免疫荧光检测重组CD317表达。用流感病毒PR8感染稳定表达重组人CD317;MTT法检测细胞活力并评价其对流感病毒的抗性作用,收集感染细胞提取RNA,用定量RT-PCR检测流感病毒在细胞中的载量。结果:免疫荧光结果表明挑取抗性细胞克隆稳定表达重组人CD317;CCK-8检测显示PR8感染后,CD317活性高于对照的空载体表达细胞。定量RT-PCR表明,PR8在该稳定表达CD317的细胞中基因载量高于空载体细胞。结论:稳定表达重组人CD317可抵抗流感的破坏作用,提示CD317可抑制PR8释放。
