银杏内酯注射液对大鼠急性期脑缺血再灌注损伤的保护作用 被引量:20

The neuroprotective effects of ginkgolide on acute cerebral ischemia /reperfusion injury in rats

摘要 目的:研究银杏内酯注射液对大鼠急性期缺血再灌注脑损伤的保护作用。方法:采用Longa法制备大鼠暂时性大脑中动脉阻塞(t MCAO)模型,于缺血1.5 h再灌后1 h给予银杏内酯注射液(0.625,1.25和2.5 mg/kg,i.p.,bid)及阳性药依达拉奉注射液(3.0 mg/kg,i.p.,bid),再灌注72 h后通过TTC染色、神经功能评分及脑水肿测定观察银杏内酯注射液对再灌注损伤的保护作用,应用免疫组化观察药物对主要神经血管单元组分神经元、星形胶质细胞、小胶质细胞和血管形态和数量的改变。结果:银杏内酯注射液(2.5 mg/kg,i.p.,bid)显著改善t MCAO模型大鼠急性期神经功能障碍、减少脑梗死体积、降低脑水肿程度,并且能调节神经血管单元组分,表现为减轻缺血再灌后大鼠半影区皮层神经元的丢失,抑制星形胶质细胞和小胶质细胞的增殖活化,降低血管外基质胶原蛋白Ⅳ(Collagen IV)的降解。结论:银杏内酯注射液对t MCAO模型大鼠急性期脑缺血再灌注损伤具有神经保护作用。 AIM: To investigate the neuroprotective effect of ginkgolide on acute cerebral ischemia / reperfusion( I / R) injury in rats. METHODS:The present study was carried out to investigate the effect of ginkgolide on acute injury in the rat I / R model induced by a 90 min temporary middle cerebral artery occlusion( t MCAO) using Longa's method. The rats were treated with ginkgolide 0. 625,1. 25 and 2. 5 mg / kg( i. p.,Bid) and edaravone3. 0 mg / kg( positive durg,i. p.,Bid) after t MCAO. Neurological deficit score,2,3,5-triphenyltetrazolium chloride( TTC) staining,brain water content,and the pathology of brain tissue( Neu N,GFAP,Iba-1 and collagen IV) were measured at 72 h after reperfusion. RESULTS: The treatment with2. 5 mg / kg ginkgolide significantly improved neurologic impairment,decreased the volume of cerebral infarction and reduced cerebral edema. Moreover,it could inhibit the loss of neurons,the proliferation and activation of astrocytes and microglia,and the degradation of collagen IV in cortex after reperfusion. CONCLUSION: These findings demonstrate that ginkgolide exhibit neuroprotective properties on acute I / R injury in t MCAO rats.
