摘要 自身免疫性疾病由于致病机制复杂且仍未探索清楚,因此临床常规药物治疗仍以免疫抑制剂为主。然而,在自身免疫性疾病的发生、发展过程中,免疫细胞上的ABC转运蛋白表达调控异常会影响初次使用的免疫抑制剂在免疫细胞内的代谢处置,造成原发性耐药;此外,随着药物治疗的持续,免疫抑制剂逐渐诱导免疫细胞上ABC转运蛋白的表达,导致进入免疫细胞的药物量减少,作用减弱,造成继发性耐药。上述两方面原因给自身免疫性疾病的药物治疗带了巨大挑战。因此,本文将从ABC转运蛋白与免疫细胞、自身免疫性疾病、药物治疗的相互影响、交互调控的角度展开阐述,为该类疾病的药物治疗提供新思路。 Due to the complicated pathogenic mechanisms of autoimmunity diseases,immunosuppressants are often used in clinical. However,abnormal regulations of ABC transport proteins in immune cells occurred with the development of the autoimmunity disease and led to drug resistance in initial drug treatment. Besides,after long-term usage of immunosuppressants,the ABC transport proteins are up-regulated and immunosuppressants could be effluxed out of the target immune cells. Therefore,the treatment of autoimmunity disease is faced with huge challenges due to these mentioned problems above. In this paper,ABC transport proteins,immune cells,autoimmunity diseases and drug therapy as well as the interactions between one and others will be reviewed,which is beneficial to provide new ideas for drug treatment of autoimmunity diseases in future.
