摘要
目的观察健康青年和老年人外周血单个核细胞(PBMC)中干扰素诱导蛋白1(IFIT1)和3(IFIT3)m RNA表达,为临床个体化治疗和疾病预防提供理论基础。方法选择373例健康人,取抗凝全血,实时荧光定量PCR检测IFIT1和IFIT3基因m RNA表达。全自动生化分析仪检测肝功能(ALT和AST)。结果健康青年女性IFIT1和IFIT3表达量与老年女性比较,差异均有统计学意义(t=11.92、14.93,P<0.01);健康青年男性IFIT1和IFIT3表达量与老年男性比较,差异均有统计学意义(t=16.64,9.15,P<0.01)。健康老年女性患者IFIT1和IFIT3表达量与老年男性比较,差异均有统计学意义(t=8.78、4.43,P<0.01);健康青年女性IFIT1和IFIT3基因表达量与青年男性比较,差异均有统计学意义(t=7.92、8.95,P<0.01)。不同年龄和性别健康人群的肝功能间进行比较,差异无统计学意义(P>0.05)。对IFIT1和IFIT3基因m RNA表达量与年龄、性别和肝功能进行相关性分析,结果仅发现IFIT1和IFIT3基因表达量与年龄、性别均呈正相关(rIFIT1=0.360、0.302;rIFIT3=0.373、0.334,均P<0.05)。结论性别和年龄与IFIT1和IFIT3基因的表达具有相关性,可为临床个体化治疗和基础研究提供新的思路。
[Objective]To observe the expression of interferon-induced protein with tetratricopeptide repeats 1 and 3(IFIT1,IFIT3)m RNA in peripheral blood mononuclear cells(PBMC) of healthy elderly and young populations,and provide a theoretical basis for clinical individualized treatment and disease prevention. [Methods]The anticoagulant whole blood was collected from 373 healthy individuals. Expression levels of IFIT1 and IFIT3 m RNA were detected by real-time quantitative PCR. The liver function(ALT and AST)were detected by automatic biochemical analyzer.[Results]The expression of IFIT1 and IFIT3 were significantly different between young and old females(t=11.92,14.93;P〈0.01). The expression of IFIT1 and IFIT3 were significantly different between young and old males t=16.64,9.15;P〈0.01). The expression of IFIT1 and IFIT3 were significantly different between old males and females(t=8.78,4.43;P〈0.01). The expression of IFIT1 and IFIT3 m RNA were significantly different between young females and males(t=7.92,8.95;P〈0.01),respectively. The liver functions were not significantly different among healthy people of different age and gender(P〉0.05). A correlation analysis of m RNA expression of IFIT1 and IFIT3 gene with ages,gender and liver function in healthy people,which only found that IFIT1 and IFIT3 were positively correlated with ages and gender(rIFIT1=0.360,0.302;rIFIT3=0.373,0.334;P〉0.05),respectively.[Conclusion]The age and gender has a relevant relationship with IFIT1 and IFIT3 genes,which provides new ideas for clinical individualized treatment and basic research.
