摘要
目的:建立细叶石仙桃多糖含量测定的方法,并对其提取工艺进行研究。方法:以葡萄糖为标准品,用苯酚-硫酸法于488nm波长处测定多糖的含量,并以其为考察指标,采用单因素试验和L9(34)正交试验设计筛选最佳提取工艺。结果:葡萄糖在2.015~10.075μg/m L范围内线性关系良好,线性回归方程为A=0.0498X+0.0871(r=0.9997),平均加样回收率为97.67%,RSD%=1.44%(n=6);细叶石仙桃多糖最佳提取工艺条件为:浸提时间2h,提取温度100℃,料液比1∶60,细叶石仙桃多糖的得率为43.50mg/g。结论:该方法简便、准确,优选出的提取工艺重复性好。
Objective:To establish the method for determining the content of polysaccharides in Pholidota can- tonensis, and to explore the extraction technique for polysaccharides from Pholidota cantonensis. Methods:Using glu- cose as the standard, the content of polysaccharides was measured at a wavelength of 488 nm by the phenol - sulfu- ric acid method. With the content of polysaccharides as an index, the univariate test and L9 ( 3^4 ) orthogonal design were performed to figure out the optimal extraction technique. Resuhs:A good linear relationship with a regression e- quation of A =0. 0498X +0. 0871 (r = 0. 9997)was achieved when the concentration of glucose ranged between 2. 015 and 10. 075μg/mL The average recovery rate was 97. 67% ,and the relative standard deviation was 1.44% (n =6). The optimal extraction of polysaccharide from Pholidota cantonensis was performed by 2 - hour soaking using 60 vol- umes of solvent at 100℃. The yield rate of polysaccharides from Pholidota cantonensis was 43.50mg/g. Conclusion: The established method is simple and accurate ,and the optimal extraction technique shows good repeatability.
