复方中药组分对糖尿病大鼠血浆P-选择素、sICAM-1的影响

摘要 目的:通过观察复方中药组分治疗链脲佐菌素制备的糖尿病模型大鼠血浆P-选择素、s ICAM-1的影响,探讨中药组分治疗糖尿病微血管并发症的作用机理。方法:将链脲佐菌素制备的糖尿病模型大鼠随机分为5组,每组10只,中药组分(麦冬多糖、黄连生物碱、三七总皂苷)高、中、低剂量组[0.45、0.15、0.05g/(kg·d)]、二甲双胍组[0.05mg/(kg·d)]、模型对照组[生理盐水10ml/(kg·d)]。连续灌胃4周,采用断头采血,用酶联免疫吸附法检测血浆中P-选择素、s ICAM-1的含量。结果:各组大鼠血浆中血管活性物质及细胞因子P-选择素、s ICAM-1含量均低于模型对照组。结论:中药组分可能通过影响P-选择素及s ICAM-1水平改善血管内皮损伤,减轻糖尿病微血管并发症。 Objective: To observe Chinese medicine components including ophiopogonpolysaccharide, total arasaponin and rhizoma coptidis alkaloid on soluble intercellular adhesion molecule ( sICAM-1) , P-selec-tion in blood plasma of model type 2 diabetic rats induced by STZ.To explore the treatment mechanism of Chi-nese medicine components.Methods:Prepare model diabetic rats aged 8 months by high fat,high glucose feed-ing and intravenous injection of Streptozotocin( STZ) ,and those successful model rats were randomly divided into five groups, Chinese medicine component high -dose group (0.45g/kg), medium -dose group (0.15g/kg), low-dose group (0.05g/kg), metformin (0.05mg/kg) group, model group and blank control group.Each group included 10 rats which were fed with the same volume medicines respectively for consecu-tive 4 weeks.Then we collected blood by cutting heads and tested the levels of soluble intercellular adhesion molecule ( sICAM-1) , P-selection in plasma.Results: The levels of sICAM-1 and P-selection in Chi-nese medicine components groups and metformin group were lower than those in the model group.Conclusion:Chinese medicine components may contribute to microvascular complications of type 2 diabetic rats by decrea-sing the levels of sICAM-1 and P-selection, which can damage endothelial cells of blood vessels.
