摘要
目的 :探讨人口腔鳞癌细胞系 (Tca8113和BcaCD885 )对颈淋巴结靶向性葫芦素BE聚乳酸纳米微粒 (CuBE_PLA_NP)的敏感性。方法 :用四唑盐显色法 (MTT法 )检测用不同剂量的CuBE_PLA_NP和CuBE在 1~ 8d内 8个时间点对Tca8113和BcaCD885的抗癌活性 ,计算出 2种药物在 8个时间点对 2种癌细胞的抑癌率和药物的半数抑癌率浓度。结果 :CuBE_PLA_NP和CuBE对Tca8113和BcaCD885均有强的杀伤作用 ,其杀伤效应均呈时间依赖性和剂量依赖性。结论 :经改型后的淋巴靶向CuBE_PLA_NP没有降低CuBE成份的抗癌活性 ,同时 ,由于CuBE_PLA_NP具有淋巴靶向性 ,更有利于杀灭口腔癌颈淋巴结转移灶内的癌细胞。
Objective:The aim of this study was to study cytotoxic effects of cucurbitacin BE polylactic acid nanoparticles (CuBE_PLA_NP) on human oral squamous carcinoma cell lines Tca8113 (tongue carcinoma) and BcaCD885 (buccal carcinoma).Methods:The chemosensitivity of Tca8113 and BcaCD885 to CuBE_PLA_NP and CuBE was tested using MTT assay, the inhibitory rate and 50% inhibition concentrations values (IC_50) of these two drugs against these two cancer cell lines were evaluated in 1,2,3,4,5,6,7 and 8 days with different concentrations.Results:Both CuBE_PLA_NP and CuBE showed very high anti_cancer effects on Tca8113 and BcaCD855, and the anticancer efficacy of these two drugs was dose_ and time_dependent.Conclusion:The effects of CuBE_PLA_NP on lymph nodes of metastatic cancer cells were higher than that of CuBE.\;
