摘要
为了简化丝素微球的制备方法及保证丝素微球作为药物缓释载体的安全性与结构稳定性,以丝素蛋白为原材料,采用自组装方法并调节丝素蛋白溶液浓度、冷冻温度、异丙醇添加量等条件,制备粒径在0.4~1.3μm之间的丝素微球。扫描电子显微镜(SEM)下观察制备的丝素微球呈规则的球状结构,球面光滑,粒径分布较为均匀;采用红外光谱仪(FT-IR)和热重分析仪(DTG)测试自组装后丝素微球的分子构象已发生转化,β-折叠结构含量明显增加,并具有较好的热稳定性。研究结果表明,丝素蛋白溶液浓度、冷冻温度、异丙醇添加量对丝素微球的形态结构有影响,可以通过自组装方法制备出粒径可控,结构和性能稳定的丝素微球,并有望开发为药物缓释载体。
For simplifying the preparation and ensuring the security and structural stability of silk fibroin microspheres as drug controlled-release carrier,silk fibroin was used as raw material to prepare silk fibroin microspheres with diameter of 0. 4-1. 3μm by modulating silk fibroin solution concentration,freezing temperature and isopropanol adding amount via self-assembly.Scanning electron microscopy( SEM) observation showed that the prepared silk fibroin microspheres performed regular spherality with smooth surface,and their particle size distributed evenly. Fourier transform infrared( FT-IR) and thermogravimetric( TG) analyses indicated that the molecular conformation of self-assembled silk fibroin microspheres was changed,the content of β-sheet increased obviously,and the microspheres had good thermostability. The results showed that silk fibroin solution concentration,freezing temperature and isopropanol adding amount had influence on the morphological structure of silk fibroin microspheres. The silk fibroin microspheres prepared by self-assembly method had characteristics of controllable particle size,stable structure and performance,being a promising drug controlled-release carrier.
