Study on metallothionein against aging effects by injecting mice with D-galactose

摘要 目的 观察外源性金属硫蛋白 (MT)是否具有抗体内自由基损伤及抗衰老效应的能力。 方法 采用小白鼠皮下注射D 半乳糖造成的亚急性衰老模型 ,以阿尼西坦作为阳性对照观察皮下注射不同剂量MT对小鼠在T 型游泳迷宫作业能力的影响及小鼠心、肝、脑组织中丙二醛 (MDA)含量和超氧化物歧化酶 (SOD)活性。 结果 MT可明显改善小鼠的学习记忆能力。MT在 1 4mg·Kg-1·d-1剂量时已表现出比阳性对照药物阿尼西坦 (30mg·kg-1·d-1)时更明显的治疗效应 (正确数获得率 ) ;同时MT在 7 0mg·kg-1·d-1时可明显降低MDA含量及提高SOD活性。 结论 MT作为内源性活性氧清除剂对D 半乳糖的致衰老效应有很好的对抗作用。 Objective To study whether exogenous metallothionein(MT) is effective against free radicals and anti aging. Methods The sub acute aging model was made by continuous subcutaneous injection of D galactose to mice inducing reactive oxygen species (ROS). Meanwhile the mice were given different doses of MT by injections. The superoxide dismutase (SOD) activity and the Malondialdehyde (MDA) content of the brain, liver and heart tissues were determined. Results The improvements on the T type water maze test were found obvious. MT inhibited the MDA increasing and SOD decreasing by scavenging the ROS. Conclusions This study demonstrates that metallothionein as an endogenous scavenger of ROS may play an effective role in preventing the tissue damages from ROS induced by D galactose.
