IGF-1R与TSHR在甲状腺相关眼病眼眶脂肪组织中作用 被引量:3

The role of IGF-1R and TSHR in the orbital adipose tissues of thyroid-associated ophthalmopathy

摘要 目的研究眼眶脂肪组织中IGF-1、IGF-1R与TSHR在TAO眼眶脂肪组织中的存在及定位。方法临床病例对照研究。对2009年8月至2014年2月在华西医院眼科就诊的TAO患者分为病例组,而非TAO为对照组。病例组:收集35例TAO患者行眼眶减压术后的眼眶脂肪组织;对照组:33例眼眶良性肿瘤或眶脂肪脱垂患者的眶脂肪组织。标本固定后行免疫组织化学染色,检测眼眶脂肪组织中IGF-1,IGF-1R,TSHR二三种因子的表达情况。结果IGF-1着染于脂肪细胞膜和细胞质,在TAO组主要为深褐色强阳性着染,强阳性率占75%,对照组染色主要为浅棕色弱阳性,强阳性率为50.0%;IGF-1R在细胞核中明显染色,TAO组呈深褐色强阳性,占76%,对照组主要为浅棕色弱阳性或阴性,强阳性率为42.8%;TSHR淡染于细胞膜。两组的染色阳性率差异无统计学意义(P〉0.05)。结论对比TSHR在本组TAO眶脂肪组织中表达不明显,而IGF-1及IGF-1R的表达强于正常,提示后两者可能与TAO眼眶脂肪组织的病理性增生有关。 Objective To investigate the presence and position of Insulin like growth factor-1 (IGF-1), IGF-1R and TSHR in orbital adipose tissue from patients with TAO. Methods The level of IGF-1, IGF-1R and TSHR in orbital adipose tissue: Both TAO group included 35 samples of orbital adipose tissue from patients with TAO who had undergone orbital decompression surgery within recent 5 years and control group included 35 samples from patients with benign orbital tumor or prolapse of orbital fat were measured by immunohistochemistry (IHC) to determine expression of IGF-1, IGF-1R and TSHR. Results Adipocytes of TAO group showed the strong positive reaction (the positive rate of IGF-1 was 75%, IGF-1R was 76%). In control group, the rate of IGF-1was 50.0%, IGF-1R was 42.8%. There was a statistically significant difference between both groups (P 〈 0.05). IGF-1 was mainly stained on the cell membrane and cytoplasm, while IGF-1R was stained on the cell nucleus. But the expression of THSR on both group showed no statistically significant difference (P 〉0.05). Conclusions Mature orbital adipocytes of TAO are mainly regulated by IGF-1 and IGF-1R comparing with TSHR.
