摘要
目的探讨Hedgehog通路(HH)抑制基因的PTCH蛋白在非小细胞肺癌(NSCLC)组织的甲基化表达以及其临床意义。方法应用甲基化特异性聚合酶链反应(MS—PCR)进行检测NSCLC中信号传导通路相关抑癌基因的甲基化状态。结果HH通路抑制基因在NSCLC中特异性不一,GAS基因的特异性为82.61%;HHIP的特异性为69.57%。PTCH的特异性为100.00%,差异均有统计学意义(P〈0.01)。结论HH通路抑制基因在肺腺癌和鳞癌中可能有不同的作用机制,住腺癌中可能通过旁分泌调控PTCH活性,而在鳞癌中则可能是通过自分泌方式调控通路活性。
Objective To explore the expression of PTCH protein in the Hedgehog pathway of suppressor gene and significance in methylation of non -small cell lung cancer (NSCLC) tissues. Methods To detect the methylation status of signal pathway correlated suppressor gene in NSCLC by using methyla- tion specific polymerases chain reaction (MS - PCR). Results The specificity of Hedgehog pathway sup- prcssor gene are different in NSCLC, GAS gene is 82. 61%, HHIP gene is 69. 57%, PTCH gene is 100.00% , the differences have statistical significance (P 〈0. 01 ). Conclusion HH pathway suppressor gcne may have different mechanisms in lung adenocarcinoma and squamous cell carcinoma. In adenocarci- noma, it may regulate activity of PTCH through paracrining, while in squamous carcinoma it may be regu- late activity of pathways by autocrining manner.
