摘要
目的检测泛素特异蛋白酶39(US39)基因在乳腺癌组织中的表达,探讨其表达与乳腺癌发生发展的关系。,方法选取108例配对的乳腺癌和乳腺癌癌旁组织,采用实时荧光定量聚合酶链反应(Real.timePCR)检测USP39mRNA在乳腺癌组织及癌旁组织中的表达,并结合临床病婵资料进行相关分析。结果USP39mRNA在乳腺癌组织中表达(△Ct=5.76±0.18)是癌旁组织(△Ct=7.19±0.19)的2.69倍,表达量显著高于癌旁组织,差异有统计学意义(t=2.36,P〈0.05)。结合临床病理资料统计分析表明,USP39的高表达与患者年龄、组织学分级、TNM分期、增殖细胞核抗原(Ki.67)表达明显相关(P〈0.05)。结论USP39mRNA在乳腺腺癌组织中明显呈高表达状念,USI^9的高表达与乳腺癌患者预后不良的一些临床病理指标相关,USP39在乳腺癌的发生发展t}l发挥作用。
Objective To evaluate the expression of ubiquitin specific protease 39 (USP39) mRNA in breast cancer tissues and assess its correlation with clinicopathological characteristics. Methods Real- time PCR was used to investigate the mRNA expression of USP39 in 108 cases of human breast caucer tissues and their matched paratumor normal breast tissues. The relationship between USP39 expres- siou and the clinicopathologic features was also evaluated statistically. Results The relative level of USP39 tuRN A in tumor samples ( △Ct = 5.76 ± 0. 18 ) compared with their matched paratumor normal samples ( △Ct =7. 19 ±0. 19) was 2. 69. The USP39 mRNA expression in breast cancer tissues was significantly higher than that in paratumor normal breast tissues ( t = 2. 36, P 〈 0. 05 ). Statistical analysis showed that tile high expression of USP39 mRNA was related to age, histological grade, TNM stage and proliferation cell nuclear antigen ( Ki - 67 ) expression of breast cancer ( P 〈 0. 05 ). Conclusion The expression of USP39 mRNA is significantly increased in breast cancer tissues and the high expression of USP39 is associ- ated with some factors which related to high malignant degree of breast cancer and poor prognosis of breast cancer patients. Our results suggest that USP39 may contribute to the occurrence and development of breast Callcer.
