蠲痹颗粒对兔膝骨关节炎软骨细胞凋亡的影响 被引量:2

摘要 目的探讨蠲痹颗粒对兔膝骨关节炎软骨软骨细胞凋亡的影响,揭示其治疗骨关节炎(OA)的部分作用机制,为蠲痹颗粒方治疗OA提供理论依据。方法将42只新西兰兔采用改良Hulth法建立兔膝OA模型,造模后,随机分为假手术组、模型组、蠲痹颗粒组及阳性药(双醋瑞因)组。药物干预或灌生理盐水6周后,取出关节软骨,石蜡切片HE染色观察软骨组织镜下病理形态改变,免疫组化法检测软骨组织caspase-3表达水平,TUNEL法检测软骨细胞凋亡情况。结果蠲痹颗粒组和阳性药对照组兔膝关节软骨组织caspase-3蛋白的表达、软骨细胞凋亡率明显低于模型组,比较有统计学意义(P<0.05)。结论蠲痹颗粒能下调caspase-3蛋白表达,抑制膝关节软骨细胞凋亡,可能是蠲痹颗粒治疗骨关节炎的作用机制之一。 Objective To explore the influence of Juanbi granule on chondrocyte apoptosis of rabbit knee osteoarthritis, reveal partial treat mechanism of the osteoarthritis(OA), and provide the theory basis for treatment of OA. Methods 42 New Zealand rabbits were used to establish improved Hulth model of knee OA. After the mould, the rabbits were randomly divided into control group, model group, Juanbi granule group and the positive drug group(double vinegar). After six weeks intervention of drugs or physiological saline, the articular cartilage was removed. We observed the pathological morphological changes of HE dyeing cartilage tissue under the mirror, detected the caspase-3 expression level in chondrocytes by immunohistochemical method, and detected chondrocytes apoptosis by TUNEL method.Rusults The rabbit knee joint cartilage caspase- 3 protein expression level and cartilage cell apoptosis rate of Juanbi granule group and positive medicine control group were significantly lower than those of the model group, and there was statistical significance(P〈0.05).Conclusion Juanbi granule can cut the caspase-3 protein expression and cell apoptosis.This may be one of the mechanisms of Juanbi granule in treating osteoarthritis.
