寻常型银屑病皮损特异性miRNA表达与靶基因研究及竹黄颗粒剂对其的干预作用 被引量:3

摘要 目的:筛选寻常型银屑病特异性miRNA并分析其靶基因及基因功能,同时观察中药竹黄颗粒剂的干预作用。方法:采集银屑病组和正常组皮肤,运用miRNA基因芯片检测两组miRNA的表达谱,分析得出差异表达的miRNA,并预测其靶基因及进行功能富集分析。同时分析竹黄颗粒剂干预前后患者皮损miRNAs的表达差异。结果:银屑病组有55个明显上调的miRNA,12个下调miRNA,其中5个特异性miRNAs靶基因预测得出AIRE等33个关键基因;功能分析得出其调控了MAPKs等30项主要信号通路;竹黄颗粒剂对银屑病皮损中miR-192等11个差异miRNA有显著影响。结论:miR-31等67个特异性miRNA可能通过影响信号通路参与了银屑病的发病,其有望成为银屑病早期诊断的潜在标记物和治疗靶点;中药竹黄颗粒剂对部分差异miRNA有双调作用,为中医药防治银屑病提供了客观依据。 Objective: Screening of psoriasis vulgaris specific miRNAs, investigating their target genes and gene function and observing the intervention of Zhuhuang Granule. Methods: Skin samples were collected from the psoriasis group and the normal group. The miRNA microarray technique was used to detect the expression profiles of two groups of miRNA. The obtained differentially expressed miRNA were analyzed. We predict the target genes and their functional enrichment analysis has been done. The effect of Zhuhuang Granule before and after the intervention in patients lesions on differentially expression of miRNA was also analyzed. Results: 55 significantly increased miRNA and 12 down-regulated miRNA were observed in psoriasis group, among them 5 specific miRNA target genes predicted 33 key genes such as AIRE etc., which regulated other 30 major signaling pathway including MAPKs using functional analysis. Zhuhuang Granule have a significant effect on 11 difference miRNAs including miR-192 in the psoriatic lesions. Conclusion: 67 specific miRNA, such as miR-31, may be involved in signaling pathways by affecting the pathogenesis of psoriasis, which is expected to become the early diagnosis of psoriasis potential markers and therapeutic targets; medicine Zhuhuang Granule have two-tone effect on partial differential miRNAs, which provide an objective basis for the TCM prevention and treatment of psoriasis.
