针刺对心肌缺血再灌注损伤大鼠心肌组织CaMK Ⅱ表达及细胞凋亡的影响 被引量:22

Influence of acupuncture on CaMKⅡ expression and apoptosis of rats with myocardial ischemia reperfusion injury

摘要 目的:观察电针手厥阴经内关、郄门穴在缺血再灌注损伤过程中对心肌组织内Ca MKⅡ蛋白含量及心肌细胞凋亡的影响。方法:将Wistar大鼠随机分为假手术组、缺血-再灌注模型Ⅰ组、缺血-再灌注模型Ⅱ组、针刺内关治疗组、针刺郄门治疗组、针刺合谷对照组、Ca MKⅡ抑制剂组(KN-93),除假手术组外,其余各组制备缺血-再灌注模型,并接受电针治疗,摘取心脏;荧光免疫组化法观察Ca MKⅡ蛋白的表达,TUNEL法观察心肌细胞凋亡率。结果:针刺内关治疗组及针刺郄门治疗组Ca MKⅡ平均光密度值高于假手术组而低于缺血-再灌注模型Ⅰ组(P<0.01),心肌细胞凋亡率显著低于两模型组(P<0.01);针刺郄门治疗组和KN-93抑制剂组Ca MKⅡ光密度值和心肌细胞凋亡率与针刺内关治疗组比较无显著差异。结论:针刺在减轻Ca2+超载过程中,心肌细胞凋亡的抑制作用与Ca MKⅡ信号通路密切相关。 Objective: To observe the protein expression of CaMK Ⅱ and myocardial apoptosis in cardiac muscle tissue of rats with myocardial ischemia reperfusion injury by acupuncture in Neiguan (PC6) and Ximen (PC4) points. Methods: Wistar rats were randomly divided into sham-operated group, model Ⅰ group, model Ⅱ group, Neiguan group and Ximen group, Hegu (LI4) control group and KN-93 group. Rats in all the groups except sham-operated group were received ischemia-reperfusion surgery and electro-acupuncture treatment. The expression of CaMK Ⅱ and myocardial apoptosis were observed by using fluorescent immunohistochemical method and TUNEL method respectively. Results: The average optical density of CaMK Ⅱ protein in rats in Neiguan and Ximen group were significantly higher than that of sham-operated group and lower than that of model Ⅰ group (P〈0.01). The myocardial apoptosis of rats in Neiguan group was significant lower than that of model Ⅰ and Ⅱ group (P〈0.01). There was no significant differences on the average optical denisity of CaMK Ⅱ and myocardial apoptosis among Ximen group, KN-93 group and Neiguan group. Conclusion: The relations between inhibition of myocardial apoptosis and CaMK Ⅱ signal path are closely in the course of calcium overload remission treated with acupuncture.
