中药方剂对牙周炎大鼠牙周组织影响研究

Influence of prescriptions of traditional Chinese medicine on periodontal tissue of periodontitis rats

目的:观察中药方剂补肾坚骨汤对牙周炎大鼠牙周组织的影响,探讨其对牙周炎的作用。方法:雄性12月龄SD大鼠24只,8只为正常组,其余成功建立大鼠牙周炎模型之后,分为牙周炎模型对照组、补肾坚骨汤治疗组16g·kg-1·d-1,给予补肾坚骨汤灌胃4周,每日1次,每次5m L,各组大鼠末次给药后24h处死,摘取上颌骨制作牙体牙周组织联合切片,HE染色,在光学显微镜下观察牙周的组织学改变;并进行骨计量学观察。结果:组织学观察,牙周炎模型对照组牙周组织表现为明显的炎性破坏,牙槽骨吸收明显,而补肾坚骨汤治疗组牙周组织炎性反应明显减轻,牙槽骨吸收减少;骨计量学观察,牙周炎模型对照组大鼠牙周结缔组织附着丧失量、牙槽骨嵴高度丧失量显著高于正常组,补肾坚骨汤治疗组大鼠牙周结缔组织附着丧失量、牙槽骨嵴高度丧失量与牙周炎组大鼠差异无统计学意义;牙周炎模型对照组及补肾坚骨汤治疗组大鼠破骨细胞数显著高于正常组,补肾坚骨汤治疗组大鼠成骨细胞数显著高于牙周炎组。结论:补肾坚骨汤可以减轻牙周炎大鼠牙周组织炎性反应,对牙周炎有一定的治疗作用。

Objective： To observe the influence of Bushen Jiangu Decoction on periodontal tissue of periodontitis rats, and investigate its therapeutic effects on periodontitis. Methods： Twenty-four 12-month-old male SD rats were randomly divided into control group, model group and Bushen Jiangu Decoction group, and with 8 rats in each group. Rats in Bushen Jiangu Decoction group received gavage administration with 5mL Bushen Jiangu Decoction （16g·kg-1·d-1） once a day for 4 weeks. All the rats were sacrificed after 24h of final administration, and the upper jaw bone was obtained to make sections of tooth and periodontal tissues. The changes in periodontal tissue were observed by using HE staining method and bone histomorphometry observation. Results： The results of histological observation were as follows： the periodontal tissue of rats in model group manifested as obvious inflammatory injury and reduced alveolar resorption. The results of bone histomorphometry observation were as follows： the lost quantities of connective tissue attachment in periodontium and alveolar ridge height of rats in model group were more than that of rats in control group, and the difference in which between model group and Bushen Jiangu Decoction group had no statistical significance; the number of osteoclasts in rats of model group was significantly more than that of control group, and the number of osteoblasts in rats of Bushen Jiangu Decoction group was significantly more than that of model group. Conclusion： Bushen Jiangu Decoction could relieve the inflammation of periodontal tissue, and has a certain therapeutic effects on periodontitis.
