不同电针频率、时程预处理对大鼠心肌缺血预适应的影响

Effects of Electro-acupuncture Pretreatment at Different Frequency and Duration in Myocardial Ischemic Preconditioning of Rats

目的:以不同电针频率、时程预处理大鼠,探讨总结诱导心肌缺血耐受的最佳电针预处理方案。方法:60只健康成年雄性Wistar大鼠,随机分为正常组、模型组、电针低频3、7天组、中频3、7天组、高频3、7天组。应用异丙肾上腺素诱导心肌缺血模型;低频电针组均给予2Hz低频电流针刺心俞穴,中频电针组予50Hz,高频电针组均予100Hz,应用生理记录仪监测心率、J点差值和收缩压情况,全自动生化分析仪测定血清CK、CK-MB、LDH含量。结果:低频7天、中频7天、高频3天和7天组均可以使心肌缺血大鼠心率、J点差值和收缩压下降,与模型组比较,差异均有统计学意义(P<0.05);高频7天组心率、J点差值和收缩压下降最显著,与低频、中频、高频3天组比较,差异均有统计学意义(P<0.05)。低频3天、7天组、中频3天、7天和高频3天、7天组血清CK、CK-MB含量均低于模型组,差异有统计学意义(P<0.05),低频7天组和中、高频3天、7天组血清LDH含量均低于模型组,差异有统计学意义(P<0.05)。高频7天组血清CK、CK-MB、LDH含量均低于低频3天和高频3天组,差异有统计学意义(P<0.05)。结论:给予高频电针心俞穴连续7天预处理,可以较好的诱导随后发生的急性心肌缺血预适应,发挥心肌保护作用。

Objective： To explore the effects of electro- acupuncture pretreatment at different frequency and duration in myocardial ischemic preconditioning of rats,to summarize the best project of electro- acupuncture pretreatment. Method： 60 Healthy male Wistar rats,were randomized divided into normal group,model group,low frequency electro- acupuncture for 3 and 7 days groups,middle frequency for 3 and 7 dnys groups,high frequency for 3 and 7 days groups. Models were induced by isoprcnalinc. Low frequency electro- acupuncture group was treated by electro- acupuncture of 2Hz,middle frequen was treated by 50 Hz,high frequency was100 Hz. The heart rate,J- point margin and systolic blood pressure were determined by physiological recorder,the contents of CK,CK- MB and LDH were measured by automatic biochemical analyzer. Results： The heart rate,J- point margin and systolic blood pressure of the electro- acupuncture pretreatment of low frequency for 7days,middle for 7days and ligh frequency for 3,7 days groups were lowered,there were significant differences compared with the model group（ P〈0. 05）,the effects of high frequency for 7 days group were the most obvious.The activities of CK and CK- MB of all groups were lower than those in model group（ P〈0. 05）,but the activity of LDH in low frequency for 7 days group and middlel,high frequency for 3 and 7 days groups were lower than that in model group（ P〈0. 05）,the activities of CK,CK- MB and LDH in high frequency for 7 days group were the lowest compared with the others（ P〈0. 05）. Conclusion： High frequency electro- acupuncture pretreatment for 7 days can protect myocardial ischemia very well.
