丹贝益肺方对肺纤维化大鼠血清TNF-α、IL-6表达的影响

Effects of Danbei Yifei Fang(丹贝益肺方) on Serum TNF-α and IL-6 Levels of Pulmonary Fibrosis Rats

目的:观察丹贝益肺方对肺纤维化大鼠血清TNF-α、IL-6水平的影响。方法:选用108只Wistar大鼠,随机分为正常对照组,模型组,丹贝益肺低、中、高剂量组,阳性药对照组。采用经气管注入博莱霉素法建立肺纤维化大鼠模型。在造模第2日起分别用生理盐水,丹贝益肺低、中、高剂量水煎液,强的松水溶液灌胃,造模后分别于第7、14、28天随机处死大鼠取材,采用酶联免疫法测定血清TNF-α、IL-6水平;HE染色及Masson染色观察大鼠肺组织炎症及纤维化程度。结果:丹贝益肺方与强的松均可明显改善大鼠的肺泡炎症改变及纤维化程度。丹贝益肺方高、中、低剂量组、强的松组血清中IL-6、TNF-α水平在各时间段均低于模型组,均具统计学意义(P<0.05),且高剂量组略低于强的松组。结论:丹贝益肺方可降低肺纤维化大鼠血清中TNF-α和IL-6的表达,抑制肺纤维化的进展。

Objective： To observe the effects of Dan Bei Yifei Fang on serum TNF- α and IL- 6 levels in pulmonary rats. Methods： 108 healthy Wistar rats were choosen,models were made by bleomycin instillation.All the rats were randomly divided into six groups,normal control group,model control group,low,moderate,high dose Danbei Yifei Fang groups,positive medicine control group（ prednisone）,on the second day of modeling,each group was given the corresponding saline or medicine,then,on the 7th day,14 th day,28 th day,rats were randomly killed,serum TNF- α and IL- 6 were determined with ELISA,the severity scores of alveolitis and fibrosis were recorded by HE and Masson stain. Results： Danbei Yifei Fang and prednisone could improve the severity of alveolitis and fibrosis. Danbei Yifei Fang and prednisone solution could lower the levels of serum TNF- α and IL- 6,the effects of high dose Danbei Yifei Fang group were obviously better than those of prednisone group. Conclusion： Danbei Yifei Fang can decrease the expressions of TNF- α and IL- 6 to ease pulmonary fibrosis of rats.
