摘要
利用DNA测序与PCR-RFLP方法,分析瓢鸡溶质载体24家族A5成员(SLC24A5)基因多态性,并与肤色性状进行了关联分析。结果表明,在SLC24A5基因第4外显子发现G482C多态位点,引起161位苏氨酸变为丝氨酸(T161S),建立了引入酶切位点的错配Nhe I PCR-RFLP分子标记,利用该标记对52只白肤色与44只黑肤色瓢鸡个体进行了基因分型。试验结果提示,SLC24A5 NheI PCR-RFLP标记与肤色性状具有显著的相关性(P<0.05),G等位基因在白肤色群体和黑肤色群体均为优势基因,黑肤色群体的C等位基因频率显著高于白肤色群体。
To investigate the correlation between polymorphism of SLC24A5 gene and skin color in Piao chicken,DNA sequencing and PCR-RFLP techniques were applied to detect SNP G482C sited in exon 4 of SLC24A5 gene and establish Nhe I PCR-RFLP genetic marker mismatch. Fifty two normal skin chicken and forty four black skin chicken were genotyped using this genetic marker. Results showed a significant corelation was detected between SNP G482C of SLC24A5 gene and skin color in Piao chicken. G allele was the dominant allele in both black skin and white skin populations, while C allele frequency in black popula- tion was much higher than that in white skin population.
