摘要
柱色谱分离荧光黄和碱性湖蓝BB是基础有机化学实验教学中的重要内容,常用带玻璃塞的色谱柱以及柱子底部使用脱脂棉进行分离。文章通过对经典色谱柱的改进,选用了新的样品溶剂和洗脱剂,改玻璃塞为四氟阀门塞,以PPC代替脱脂棉,借助分光光度法绘制它们的标准曲线,定量测定洗脱以后的浓度,从而计算它们各自的收率,收率分别为98.1%和97.8%,对教材实验操作由定性分析转向定量分析,有很好的分离效果。
Column chromatography separation of fluorescent yellow and methylene blue BB is an important part of the foundation of Organic Chemistry Experimental Teaching. Through the improvement of the classical column chromatography, by means of spectrophotometry plotted the standard curve to calculate their yield, the yield was 98.1%and 97.8%, experimental operation turned from qualitative analysis to quantitative analysis, the separation effect was good.
