基质细胞衍生因子-1及Wnt信号对肌成纤维细胞生物特性的影响

摘要 目的探讨基质细胞衍生因子-1(SDF-1)及Wnt信号通路对诱导生成的肌成纤维细胞的生物学特性的影响。方法首先体外诱导骨髓间充质干细胞(BMSCs)分化为肌成纤维细胞(MF),然后改变SDF-1及(或)Wnt信号,通过划痕实验观察MF迁移能力的变化,qRT-PCr检测Ⅰ型胶原(ColⅠ)、α-平滑肌肌动蛋白(α-SMA)表达的改变。结果转化生长因子(TGF-β)及高浓度血清等条件的诱导,可使BMSCs分化为MF。阻断SDF-1或Wnt信号时,会显著抑制MF的迁移及Col-Ⅰ的分泌,阻断Wnt信号可明显抑制BMSCs向MF的分化。结论 SDF-1与Wnt信号参与调节MF的迁移及ColⅠ分泌,Wnt信号对BMSCs向MF分化具有重要作用。 Objective To explore the impact of SDF-1 and wnt signal pathway on the biological characteris-tics of Myofibroblast .Methods Firstly the BMSCs were induced to differentiate into MF,then after the change of SDF-1 and (or)Wnt signal pathway ,the MF's type Ⅰ collagen(ColⅠ)and α-smooth muscle actin(α-SMA)mRNA expression were detected and the migration ability of MF was evaluated by scratch experiments.Results (1)BM-SCs could be induced to differentiation into MF in the condition of culture medium including transforming growth fac-tor-β(TGF-β)and high concentration of serum in vitro;(2)When SDF-1 /CXCR4 or Wnt signal pathway were blocked ,the migration ability of MF and secretion of ColⅠwas inhibited significantly;(3)The BMSCs differentiation into MF can be inhibited by blocking Wnt signal pathway.Conclusion SDF-1 and Wnt signal pathway play an im-portant role in the regulation of MF migrationand ColⅠ secretion ,and Wnt signal pathway involves in the regulation of BMSCs differentiation into MF.
