瑞舒伐他汀对小鼠血管平滑肌细胞CXCR2表达及趋化作用的影响

摘要 【目的】探讨瑞舒伐他汀对体外培养小鼠血管平滑肌细胞CXC趋化因子受体2(CXCR2)表达及其介导的趋化作用的影响。【方法】体外分离培养原代小鼠颈动脉平滑肌细胞,分别以不同浓度氧化低密度脂蛋白(ox-LDL)和干扰素-γ(IFN-γ)诱导,免疫组化和ELISA检测瑞舒伐他汀预处理对IFN-γ诱导原代血管平滑肌细胞CXCR2表达的影响。Transwell小室实验分别检测白细胞介素-8(IL-8)单独干预或者联合瑞舒伐他汀对血管平滑肌细胞趋化作用的影响。【结果】IFN-γ和OX-LDL干预后血管平滑肌细胞CXCR2表达明显增加(P〈0.05)。瑞舒伐他汀预处理能够显著抑制IFN-γ诱导血管平滑肌细胞CXCR2表达能力,IL-8能够增强血管平滑肌细胞的趋化能力,而瑞舒伐他汀能显著抑制这种趋化作用。【结论】瑞舒伐他汀可抑制小鼠血管平滑肌细胞CXCR2表达及其介导的趋化作用。
