闽产石仙桃无菌播种与组培快繁技术研究初报 被引量:5

摘要 为构建石仙桃无菌播种与组培快繁技术,对石仙桃的无菌播种与快繁技术进行了相关研究。结果表明:石仙桃种子最佳消毒溶液为0.1%升汞,处理时间为8min;种子萌发最佳基本培养基为N6培养基,N6+1.0mg·L-1 6-BA+1.0mg·L-1 NAA激素组合的培养基对种子萌发也同样合适;不同植物激素组合对石仙桃幼苗生长影响无显著性差异,N6+1.5mg·L-1 6-BA+0.5mg·L-1 NAA效果较好;在N6+0.5mg·L-1 6-BA+1.5mg·L-1 NAA+75g·L-1香蕉培养基上生根率达85%,通过植物组织培养技术,成功构建了石仙桃种子的非共生萌发和快繁技术体系。 In order to build the seed rapid propagation technology of Pholidota chinensis, the aseptic seeding and tissue culture of Pholidota chinensis were studied. The results showed that the disinfeetion treatment with 0. 1% HgCl2 solution for 8 min was most suitable for aseptie seeding. The best medium for seed germination was N6 medium,the optimum medium of N6 +6-BA 1.0 mg·L^-1 +NAA 1.0 mg·L^-1 was also suitable for seed germination; Different combinations of 6-BA and NAA didfft show significant difference on the growth of young seedlings of Pholidota chinensis, and the appropriate medium was N6+ 6-BA 1.5 mg·L^-1 + NAA 0.5 mg·L^-1. Seedling's rooting rate was 85% on the medium of N6+6-BA 0.5 mg·L^-1+NAA 1.5 mg'L-1 +75 g·L^-1 banana. The system of asymbiotie seed germination and rapid propagation of Pholidota chinensis was constructed sueeessfully by the technology of plant tissue culture.
