摘要
目的:研究复脉汤对缺血性心律失常作用机制与依赖于MAPK信号的Cx43 Ser262磷酸化通道介导有关。方法:将SD大鼠分为假手术组,模型组,酒石酸美托洛尔组,炙甘草汤组,复脉汤高、中、低剂量组,灌胃给药14 d后,结扎SD大鼠冠状动脉左前降支(除假手术组外),制成缺血性心律失常模型,术后采用蛋白免疫印迹法(Western blot)测定各实验组大鼠心肌组织细胞外信号调节的蛋白激酶(ERK1/2)表达量及Cx43 Ser262磷酸化水平。结果:复脉汤各剂量组、美托洛尔组均可不同程度的降低ERK1/2及Cx43 Ser262磷酸化水平相对表达量,从而对缺血性心律失常起到保护作用。结论:复脉汤对缺血性心律失常的保护作用可能基于ERK1/2表达量减少,其介导的Cx43 Ser262磷酸化水平降低,维持缝隙连接蛋白结构,维系缝隙通讯功能。
Objective: To research the mechanism of Fumai decoction using on ischemic arrhythmias depended on Cx43 Ser262 phosphorylation channel that mediated by MAPK signal. Methods: The experiment is divided into Sham operation group, Model group, Metoprolol tartrate group, Zhigancao decoction group, high, medium and low dose groups of Fumai decoction. After the administration in 14 days, SD rat models of ischemic arrhythmia is producted by ligating the left anterior descending artery ( except sham group ) .Then, We determinate expression of extracelluar signal-regulated protein kinase ( ERK1/2 ) and phosphorylation expression of Cx43 Set 262 by Western blot in myocardial tissue. Results : Each dose group of Fumai decoction and metoprolol group could reduce the expression of ERK1/2 and Cx43 Ser262 phosphorylation and play a protective role in ischemic arrhythmias. Conclusion : Fumai decoction has the effection of protection ischemic arrhythmias based on maintaining Connexin structure and communication gap function by reducing expression of ERK1/2 and Cx43 Ser262 phosphorylation that mediated by ERK1/2.
