瑞舒伐他汀对大鼠急性心肌梗死后致命性心律失常和心功能的影响 被引量:5

Effects of rosuvastatin on fatal cardiac arrhythmias and cardiac function in rats with acute myocardial infarction

摘要 目的探讨不同剂量的瑞舒伐他汀对大鼠急性心肌梗死后心律失常和心功能的影响。方法通过结扎大鼠冠脉前降支制备大鼠急性心肌梗死模型,将造模成功并且术后24 h存活的SD大鼠45只随机分为心梗组、小剂量瑞舒伐他汀组(小剂量组)、大剂量瑞舒伐他汀组(大剂量组),每组15只。另取15只作为假手术组,开胸在相同位置挂线不结扎。小剂量组给予1 mg/(kg·d)瑞舒伐他汀、大剂量组给予10 mg/(kg·d)瑞舒伐他汀连续灌胃1周,假手术组和心梗组给予相同体积蒸馏水灌胃。1周后应用超声心动图测定各组心脏结构和功能,多导电生理仪测定血流动力学参数,程序性刺激行在体电生理检测,使用ELISA试剂盒测定各组术前和术后血脂及BNP水平。结果与假手术组比较,心梗组的QTc间期明显延长,室颤阈值下降,室性心律失常诱发率高,并且心脏收缩和舒张功能受损,BNP水平显著性升高,差异均有统计学意义(P均<0.05)。与心梗组相比,小剂量组和大剂量组QTc间期缩短,室颤阈值提高,室性心律失常诱发率下降,同时改善急性期心功能,BNP也明显下降(P均<0.05)。但各组间血脂水平比较差异无统计学意义(P均>0.05)。结论瑞舒伐他汀能有效降低大鼠心肌梗死急性期的室性心律失常诱发率,提高室颤阈值和改善心功能。 Objective It is to investigate the influence of different doses of rosuvastatin on cardiac arrhythmias and cardiac function in the acute phase of myocardial infarction in rats. Methods The rat acute myocardial infarction model was made by ligation of the anterior descending coronary artery in rats. The successful models of 45 SD rats were randomly divided into MI group (group B) , small dose rosuvastatin group (group C) , high dose rosuvastatin group (group D) , and 15 cases in each. Another 15 rats were served as sham operation controls (group A). The animals were given rosuvastatin in does of 1 mg/(kg · d) in group C and 10 mg/( kg ·d) in group D by continuous administration for a week, and equal dose distilled water was giv- en in group A and group B. Electrophysiological examination in vivo and hemodynamic examination were taken after 1 week, and the level of blood lipids and BNP were detected by ELISA. Results Compared with group A, the prevalence of ventricular arrhythmias was significantly higher in group B, with lower threshold of ventricular fibrillation, and the systolic and diastolic dysfunction and QTc interval prolongationed?, there were significant difference between the two groups ( all P 〈 0.05 ). Com- pared with group B, the prevalence of ventricular arrhythmias was significantly lower in group C and D, with higher threshold of ventricular fibrillation, and QTc interval shortened and improve heart function, there were significant differences? ( all P 〈 0.05 ). However, there was no statistical difference in lipid levels among all the groups ( all P 〉 0.05 ). Conclusion Rosuvas- tatin can effectively reduce the incidence of ventricular arrhythmias in rats with myocardial infarction, improve the heart func- tion in acute stagethe and enhance ventricular fibrillation threshold.
