摘要
为了能够快速、低成本检测油料饼粕中的真菌毒素玉米赤霉烯酮(ZEA),采用前期研制成的高灵敏、高特异性ZEA单克隆抗体2D3,经物理吸附法与纳米金颗粒偶联形成特异性探针;研究了pH、抗体标记浓度、抗体最小标记量等条件对纳米金试纸条分析灵敏度的影响,最终成功研制出ZEA免疫层析试纸条,其肉眼检测限达0.5 ng/mL;通过添加回收试验评价了该试纸条用于检测油料饼粕中ZEA方法的准确度,结果表明,供试样品的加标回收率达90%-95%;比对应用试验结果表明,实际样品检测与HPLC检测结果没有显著差异。因此,本研究提供的方法测定结果准确、重现性好,可应用于油料饼粕的现场快速筛查。
Based on the monoclonal antibody 2D3, an immunochromatographic assay was developed for rapid and low-cost determination of Zearalenone(ZEA) in oilseed meals. The method showed a limit of detection of 0. 5 ng/mL and spiked recoveries of 90% -95%. The proposed method was also compared with standard high-performance liquid chromatography (HPLC) method using 17 blind milk samples, resulting in high consistency. The results demonstrated this method could accurately measure the content of ZEA in oilseed meals.
