裸花紫珠的组培快繁技术体系

摘要 为了给裸花紫珠种苗的规模化生产提供可靠的技术途径,以野生裸花紫珠茎段为外植体,采用组织培养的方法建立裸花紫珠组培快繁技术体系。结果表明:用0.1%升汞溶液消毒10min最佳,污染率小于16%,腋芽萌芽率70%;以MS为基本培养基,6-BA浓度为0.5mg/L时,外植体芽诱导率最高,为87.5%;6-BA和NAA配合使用,继代增殖最佳,繁殖周期为30d,增殖系数为10;以1/2MS为基本培养基,IBA浓度为1.0mg/L时,生根率100%,根粗壮、有须根;河沙、椰糠及红土体积比为(1-2)∶1∶1时,移栽成活率达95%以上。 Stem segments from wild C. nudiflora were cultured to build a rapid propagation technical system for large-scale production of C. nudiflora seedlings. Results:The contamination rate and auxiliary bud germination rate of stem segments treated with 0. 1% HgCl2 solution for 10 min can be 〈 16% and 70% respectively. The bud induction rate of stem segments cultured on MS + 0. 5mg/L 6-BA is the highest (87. 5%). The optimum medium for subculture multiplication is MS + 6-BA + NAA and its reproductive cycle and multiplication coefficient are 30 d and 10 separately. The rooting rate of stem segments cultured on 1/2 MS + 1.0 mg/L IBA reaches 100% and its strong root can develop more fibrous roots. The survival rate of plantlets cultured on the substrate with river sand(1-2), coco coir (1) and red soil(1) is above 95% .
