Effect of different modes of oscillation on ELISA results using the microlab FAME system

摘要 目的探讨进行ELISA实验时,不同振荡混匀方式对FAME全自动酶免系统检测结果的影响。方法选择HBs Ag试剂盒(A/B/C)、HCVAb试剂盒(a/b/c)在同一台全自动酶免系统进行匹配实验,根据振荡混匀参数不同,设置低速振荡(B/b)组和高速振荡(C/c)组,每组再根据振荡混匀时间(0、10、20、30 s)不同分为4个小组(B0~B3/b0~b3、C0~C3/c0~c3),实验另设手工充分混匀对照(A/a)组,每组采用低值质控血清作为标本进行实验。计算各组(x±s)及变异系数(CV),对最终的检测结果进行分析。结果 HBs Ag试剂B、C组结果与相应充分混匀A组比较,B0(0 s)、B1(10 s)、C0(0 s)、C1(10 s)组的结果差异有统计学意义(P〈0.05);HCVAb试剂b、c组结果与相应充分混匀a组比较,b0(0 s)、c0(0 s)组的结果差异有统计学意义(P〈0.05);振荡时间相同而振荡频率不同的B/b组和C/c组之间比较差异无统计学意义(P〉0.05)。结论振荡方式是全自动酶免系统运行的重要环节之一,其是否合理选择会直接影响ELISA实验结果的准确性。 Objective To discuss whether different modes of oscillation can affect the results of FAME automatic ELISA test. Methods According to different parameters of oscillation, selected HBsAg kit (A/B/C), HCVAb kit (a/b/c) in the same automatic ELISA system; for matching experiments, low-speed group (B/b) and high-speed group (C/c) were selected. Each group was further divided into four different groups (B0-B3/b0-b3, C0-C3/c0-c3) by different blending time (0 s, 10 s, 20 s, 30 s). In the experiment, manual group was used as control group (A/a), and low value control serum as samples. The final test results of each group were calculated and analyzed by (x±s) and coefficient of variation (CV). Results B and C groups in HBsAg reagents compared with the corresponding group A, there were statistically significant difference between group B0 (0 s), B 1 ( 10 s), CO (0 s), C 1 ( 10 s) and control (P〈0.05) ; that was the same between group a with group b0 (0 s ), c0(0 s) and control(P〈0.05) in HCVAb reagents. There were also no statistically significant difference between groups B/b and C/c (P〉0.05). Conclusion The oscillation is one of the important aspects in automatic enzyme system operation, and the reasonable choice of oscillation modes directly affect the accuracy of ELISA experimental results.
