The serological characterization and molecular background study of red cell A_3 subgroup in blood donor

摘要 目的研究A3亚型的血清学特征。方法血液标本采集后分成2份,一份做常规血清学实验,包括ABO血型定型、吸收放散实验、抗体筛选实验;另一份抽提DNA后,扩增ABO血型基因6、7外显子并进行测序研究。留取该同一来源的唾液标本进行中和抑制实验,以检测唾液中的ABH血型物质。结果献血者标本ABO血型正、反定型不一致,红细胞可吸收放散抗-A,血清未检出不规则抗体,唾液检出A、H血型物质。测序结果显示其第6外显子C:297A>G(杂合),第7外显子C:474C>T(纯合)。结论该献血者为A3亚型,发现1个新的突变位点C:474C>T。 Objective To study the serological characterization and molecular background of A3 subgroup blood sample. Methods The blood sample was divided into 2 parts; one was used to do the routine serological tests and the other was used to do sequencing test. The donor's saliva was also collected to detect the ABH antigen. Results The donor's sense reaction of ABO blood group wasn't same as to its reverse reaction; antibody A could be absorbed by the blood sample and be eluted; antigen A and H were also detected in the donor's saliva sample. Sequencing results showed that there were two mutations, C: 297A〉G in exon 6 and C:474C〉T in exon 7. Conclusion The donor's ABO blood group is A3 subgroup, and C:474C〉T in exon 7 is a new mutation.
